Dexamethasone(MK 125; NSC 34521; Prednisolone F) is a potent synthetic member of the glucocorticoid class of steroid drugs that has anti-inflammatory and immunosuppressant properties.
DEX in concentrations from 1 to 100μM did not change LDH release but exposure to 10μM and 100μM DEX for 24, 48 and 72h caused a significant reduction in cell viability and proliferation as confirmed by MTT reduction and BrdU ELISA assays, respectively. Dex impaired neuron and oligodendrocyte maturation in PUFA-supplemented cells, demonstrated by the reduction of neurite lengths and oligodendrocyte sizes.
in vivo: The levels of IL-17, IL-6 and Tim-3 were substantially increased and the IL-10 level decreased in BALF in the asthmatic mice, which was significantly reversed by DEX treatment.An IM bolus injection of DEX sodium phosphate in pigs resulted in a fast and complete absorption, with a mean maximal plasma concentration of 80.94±21.29ng/mL at 0.35±0.21h and a high absolute bioavailability of 131.06±26.05%.
