NLG-8189 is an inhibitor of the enzymes and indolamine 2,3-dioxygenase 1 (IDO) and indolamine 2,3-dioxygenase 2 (INDOL1), with selectivity for INDOL1. The enzymes perform similar transformations and are responsible for catalyzing the rate-limiting step of oxidative tryptophan catabolism in the kynurenine pathway. IDO activity is correlated with an induction of tolerance and immunosuppresion through activation of regulatory T cells by metabolites generated from tryptophan catabolism. Inhibition of IDO by NLG-8189 blocks this induced immune suppression, which has shown utility in suppressing acquired immunities of tumors and indicates potential for chemical intervention of chronic inflammatory diseases.
